E-mail: marcellaradael@yahoo.com.br Farmed fishes are highly sensitive to diets deficient in ascorbic acid with clinical symptoms that affect performance and commercialization for causing deformities. Aiming to meet the minimum levels for red rainbow fish, six isoaminoacid and isocaloric diets with varying levels of active vitamin C were tested. At the end of 42 days experimental period, 36 fishes were histologically analyzed for the percentage of connective tissue and muscle fibers in order to determine the influence of the Vitamin C percentage in those ratios. Although the averages are statistically indistinguishable from the quantitative point of view, qualitatively higher levels of vitamin C allow better tissue structure. The breakdown of muscle tissue is indicative for dietary deficiency of ascorbic acid.

E-mail: marcellaradael@yahoo.com.br Peixes cultivados mostram-se altamente sensíveis a dietas deficientes em ácido ascórbico com sinais clínicos que afetam o desempenho e a comercialização por provocar deformidades. Com objetivo de conhecer os níveis mínimos para a melanotênia-maçã, foram testadas seis rações isoaminoacídicas e isocalóricas com níveis variados de vitamina C ativa. Ao final dos 42 dias experimentais, 36 peixes foram analisados histologicamente quanto à porcentagem de tecido conjuntivo e fibras musculares a fim de determinar a influência da porcentagem de Vitamina C nesses índices. Apesar de as médias serem estatisticamente iguais do ponto de vista quantitativo, qualitativamente os níveis mais altos de vitamina C possibilitaram melhor estrutura tecidual. A desestruturação do tecido muscular é um indicativo da deficiência alimentar com ácido ascórbico.

E-mail: msucupir@usp.br Horses used for the game of polo experience abrupt and frequent changes in exercise intensity. To meet this variable energy demand, the horses use both aerobic and anaerobic pathways in varying proportions and intensities. In this context, there must be a balance between the formation of reactive oxygen species (ROS) and the action of antioxidants to prevent oxidative stress and its consequences. The effect of supplementation with an ADE vitamin complex on oxidative metabolism was evaluated in 18 crossbred horses randomly divided between a treated group (TG) and a control group (CG). The TG animals received the ADE vitamin complex (1mL/50 kg of body weight) by deep intramuscular injection at 30 and 15 days before the game. The CG horses received 10ml of saline by the same administration route and schedule. During the polo match, the animals played for a total of 7.5 min. Blood samples were collected on the same days as the treatments were administered, and immediately before and at 15, 90 and 180 minutes after the game. The concentrations of creatine phosphokinase (CK), lactate dehydrogenase (LDH), lactate, glucose, aspartate aminotransferase (AST), glutathione (GSH), superoxide dismutase (SOD) and malondialdehyde (MDA) were measured in the blood samples. After the game, the TG demonstrated higher levels of AST, lactate and glucose than the CG, suggesting more efficient energy use by the treated animals. The higher GSH and lower lactate levels in the TG before the game suggest the presence of a greater antioxidant supply in the treated animals. The maintenance of the MDA levels indicates that neither of the groups exhibited oxidative stress.

Para satisfazer esta demanda inconstante de energia, os animais utilizam as vias aeróbia e anaeróbia em proporções e intensidade variáveis. Neste contexto deve haver equilíbrio entre a formação das espécies reativas de oxigênio (EROs) e a ação das substâncias antioxidantes a fim de evitar o estresse oxidativo e suas consequências. Avaliou-se o efeito da suplementação com vitaminas ADE no metabolismo oxidativo destes animais. Para tanto, 18 equinos mestiços foram distribuídos aleatoriamente em dois grupos: tratado e controle (GT) e controle (GC).Os animais do GT receberam complexo vitamínico ADE (1 mL/50 kg de peso vivo) pela via intramuscular profunda aos 30 e 15 dias antes do jogo. Os cavalos do GC receberam, pela mesma via de administração e nos mesmos momentos, 10mL de solução fisiológica. Os animais jogaram um tempo de 7,5min. Foram coletadas amostras de sangue nos mesmos dias de tratamento e imediatamente antes e aos 15, 90 e 180 minutos após o jogo. Foram determinadas as concentrações sanguíneas de CK, LDH, lactato, glicose, AST, GSH, SOD e MDA. Após o jogo o GT apresentou maiores valores para AST, lactato e glicemia que o GC, sugerindo melhor aproveitamento energético dos animais tratados. Os valores maiores de GSH e menores de lactato no GT antes da prova sugerem maior aporte antioxidante nos animais tratados. A manutenção dos teores de MDA indica que nenhum dos grupos entrou em estresse oxidativo.

E-mail: hill@vm.uff.br The increase of heart collagen fibers in diabetics is a well known fact, but the consequences are not defined. The aim was to quantify the cardiac collagen fibers in normal and diabetic rats treated with vitamin C. We selected 32 Wistar rats, 16 diabetic animals induced endovenously with streptozootocin, and 16 healthy animals, half of them, diabetics and normals, were treated with vitamin C for 90 days. After the experimental proceeding, the hearts were removed and processed accordingly to conventional protocol for optical microscopy and specific staining for collagen. The results showed that the diabetic rats presented increase in the number of cardiac collagen fibers, but the ones treated with vitamin C showed little accumulation of fibers. It could be concluded that treatment with vitamin C is important for the prevention of heart failure in diabetic animals.

E-mail: hill@vm.uff.br O aumento do conteúdo de fibras colágenas no coração de diabéticos é um fato bastante conhecido, suas conseqüências ainda são objeto de estudo e causam certa controvérsia, portanto este trabalho objetivou estudar a variação na quantidade das fibras de colágeno cardíacas em animais normais e diabéticos tratados pela vitamina C. Para isso foram selecionados 32 ratos Wistar, 16 diabéticos induzidos pela injeção endovenosa de estreptozootocina e 16 normais, sendo metade deles tratados com Vitamina C (diabéticos e normais) por um período de 90 dias. Após período experimental, os corações foram retirados e processados segundo protocolo convencional para microscopia óptica e coloração específica para colágeno. Os resultados mostram que animais diabéticos apresentam maior quantidade de fibras de colágeno cardíacas e que o tratamento com a vitamina C determinou um menor acúmulo na quantidade dessas fibras.

Histological and morphometric analyses were applied on 5µm thick samples and showed that: in the amount of germinative folicules, we observe resulted comparing, independent of the sex, animals controls supplemented with vitamin C and controls not supplemented (p£0.05; F=0.1452); in the amount of vases, we observe resulted comparing, of diabetic females supplemented by vitamin C when compared with not supplemented (pd”0.05; F=6.8893); and in the diameter of the vases, we observe resulted comparing females, as much in the group has controlled how much to the diabetic group, supplemented with vitamin C when compared with the females not supplemented with vitamin C (p<0.05; U=121.50; Z(U)=2.1234) and (p<0,05; F=4.8134). In a general way, the induction of diabetes modifies the vascular diameter in the females and that the vitamin C administration only intervenes with relative metric data to the vascular diameter in the females.

Among the theories proposed for its etiology the oxidative stress stands out. The performance of nitric oxide as a link between the metabolic and vascular neuropathogenic factors that triggers the diabetic neuropathy has already been put forward. This study aimed to assess the quantification and measurements of the cell body profile area (CBPA) of NADPH-diaphorase reactive (NADPH-dp) myenteric neurons of the jejunum of diabetic rats (induced by streptozotocin) supplemented with Ascorbic Acid (AA). These changes in the myenteric neurons seem to be related to the gastrointestinal disturbances observed in diabetes mellitus (DM). Twenty male Wistar rats (Rattus norvegicus) were distributed in 4 groups (n=5): controls (C), control supplemented (CS), diabetic (D), and diabetic suplemented (DS). DM was induced by estreptozotocin (50mg/kg body wt). One week after the induction and confirmation of the DM (glycemia exam), animals of the groups CS and DS received 50mg of AA three times a week by gavage. After 90 days of experiment, the animals were anesthetized with lethal thiopental dose (40mg/kg) and the collected jejunum processed for the histochemistry NADPH-diaphorase technique. Whole-mount preparations were obtained for quantitative and morphometric analysis of the myenteric neurons. A quantity of jejunum neurons in the Group D (96±7.5) was not different (P>0.05) from Group DS (116±8.08), C (92±9.7), and CS (81±5.4), but in Group DS the quantity was higher (P<0.05) than in Group C and CS. The CBPA of neurons from Group D (189.50±2.68µm2) and DS (195.92±3.75µm2) were lower (P<0.05) than from Group C (225.13±4.37µm2) and CS (210.23±3.15µm2). The streptozotocin-induced DM did not change the jejunum-ileum area, the jejunum myenteric plexus space organization and the density of NADPH-dp neurons. The 50g AA-supplementation, three times a week, during 90 days, did not decrease hyperglycemia; however, it had a neuroprotective effect on the myenteric neurons, minimizing the increase on the CBPA of NADPH-dp neurons and increasing the amount of NADPD-dp neurons.
